Serodiagnosis of Tuberculosis Using Absorbance-difference on Immunoblots



Abstract
This study was done to develop a serodiagonstic procedure for tuberculosis.
The sera from 40 patients with pulmonary tuberculosis who were found by chest
X-ray examination, smear and cloture and from 25 healthy persons were reacted with
antigen prepared with cell extract of Mycobacterium tuberculosis by immunoblotting.
Absorbance of the bands visulized by enzyme reaction was measured at 600 §¬ and
analyzed and the results obtained are as follow.
The specificity of the 24 kD antigen was 100% and the sensitivity, 37.5%.
The specificity of the 35 kD antigen was 96% and the sensitivity, 70%.
Comparative analysis of absorbance of 24 kD, 28 kD, 29 kD and 35 kD antigens
permits to diagnose active tuberculosis with the probability of more than 70%.
Thus when the absorbance-difference of these antigens is projected, it may be useful
for diagnosing patients who are negative on smear and culture examination.
